Abstract
Although tomato seeds (an industrial by-product) are known to contain several bioactive compounds, studies describing their health effects are limited. Previously, we evidenced that aqueous extract of tomato seeds (TSE) markedly attenuated rotenone (ROT)-induced oxidative stress and neurotoxicity in Drosophila system. This study investigated the neuroprotective effect of TSE in a chronic ROT model of neurotoxicity in mice. Initially, we assessed the potential of oral supplements of TSE to modulate the levels of endogenous markers of oxidative stress in brain regions of mice. Subsequently, employing a co-exposure paradigm, the propensity of TSE (100 mg/kg bw, 3 weeks) to attenuate ROT-induced behavioral phenotype (gait abnormalities, anxiety-like state), oxidative dysfunctions and neurotoxicity was examined. We found that mice provided with TSE supplements exhibited progressive improvement in gait pattern and exploratory behavior. TSE markedly offset ROT-induced oxidative impairments,restored reduced glutathione levels, antioxidant defenses (superoxide dismutase, glutathione peroxidase) and protein carbonyls content in brain regions. Specifically, TSE effectively diminished ROT induced elevation in the activity levels of acetylcholinesterase and restored the dopamine levels in striatum. Interestingly, in mitochondria, TSE was able to restore the activity of mitochondrial complexes and redox state. Collectively, our findings in the chronic ROT model demonstrate the ability of TSE to alleviate behavioral phenotype, oxidative stress, mitochondrial dysfunction and neurotoxicity. Further studies in dopaminergic cell models are necessary to understand the precise molecular mechanism/s by which tomato seed bioactives offer significant neuroprotection.–PMID—24831121–[PubMed – as supplied by publisher]

Date: May 30, 2014 Source: University of Michigan Health System Among men treated for prostate cancer, those who received radiation therapy were more likely to develop bladder or rectal cancer, according to a new study from the University of Michigan Comprehensive Cancer Center.—“Overall the incidence of these cancers is low. But when men have received radiation treatments, it’s important to evaluate carefully any symptoms that could be a sign of bladder or rectal cancer,” says senior study author Kathleen A. Cooney, M.D., professor of hematology/oncology and urology at the U-M Medical School.–The study, which will be presented at the American Society of Clinical Oncology annual meeting, looked at 441,504 men diagnosed with prostate cancer between 1992 and 2010. Men were identified from the Surveillance, Epidemiology and End Results (SEER) program, a network of National Cancer Institute-sponsored, population-based cancer registries that collect information on cancer diagnoses andtreatment. SEER performs regular follow-up for survival and to capture new invasive cancer diagnoses.–The researchers looked at the number of secondary cancers that developed 10 or more years after men were diagnosed with prostate cancer.[F1] As a whole, men diagnosed with prostate cancer were at a lower risk of developing a second cancer. But when researchers looked at patients who received external beam radiation therapy, they found these patients were estimated to be 70 percent more likely to be diagnosed with a rectal cancer and 40 percent more likely for bladder cancer than the general public.–Radiation therapy is a standard treatment for prostate cancer and the researchers stress that their findings should not prohibit anyone from choosing this treatment[F2], in particular men who are not good candidates for surgery.–“Prostate cancer has an excellent prognosis. Abstract PURPOSE: The power asymptote (critical power [CP]) and curvature constant (W’) of the power-duration relationship dictate the tolerance to severe-intensity exercise. We tested the hypothesis that dietary nitrate supplementation would increase the CP and/or the W’ during cycling exercise. METHODS: In a double-blind, randomized, crossover study, nine recreationally active male subjects supplemented their diet with either nitrate-rich concentrated beetroot juice (BR; 2 × 250 mL·d, ∼8.2 mmol·d nitrate) or a nitrate-depleted BR placebo (PL; 2 × 250 mL·d, ∼0.006 mmol·d nitrate). In each condition, the subjects completed four separate severe-intensity exercise bouts to exhaustion at 60% of the difference between the gas exchange threshold and the peak power attained during incremental exercise (60% Δ), 70% Δ, 80% Δ, and 100% peak power, and the results were used to establish CP and W’. RESULTS: Nitrate supplementation improved exercise tolerance during exercise at 60% Δ (BR, 696 ±120 vs PL, 593 ± 68 s; P < 0.05), 70% Δ (BR, 452 ± 106 vs PL, 390 ± 86 s; P < 0.05), and 80% Δ (BR, 294 ± 50 vs PL, 263 ± 50 s; P < 0.05) but not 100% peak power (BR, 182 ± 37 vs PL, 166 ± 26 s; P = 0.10). Neither CP (BR, 221 ± 27 vs PL, 218 ± 26 W) nor W' (BR, 19.3 ± 4.6 vs PL, 17.8 ± 3 kJ) were significantly altered by BR. Currently, the Herba Epimedii and its multi-component formulation are the attractive native herbs for the treatment of osteoporosis. Icariin, a typical flavonol glycoside, is considered to be the main active ingredient of the Herba Epimedii from which icariin has been successfully extracted. Most interestingly, it has been reported that icariin can be delivered locally by biomaterials and that it has an osteoinductive potential for bone tissue engineering. This review focuses on the performance of icariin in bone tissue engineering and on blending the information from icariin with the current knowledge relevant to molecular mechanisms and signal pathways. The osteoinductive potential of icariin could be attributed to its multiple functions in the musculoskeletal system which is involved in theregulation of multiple signaling pathways in anti-osteoporosis, osteogenesis, anti-osteoclastogenesis, chondrogenesis, angiogenesis, and anti-inflammation. The osteoinductive potential and the low price of icariin make it a very attractive candidate as a substitute of osteoinductive protein-bone morphogenetic proteins (BMPs), or as a promoter for enhancing the therapeutic effects of BMPs. Author information Abstract OBJECTIVES: To To compare testosterone undecanoate versus propionyl-L-carnitine plus acetyl-L-carnitine and placebo in the treatment of male aging symptoms.METHODS: A total of 120 patients were randomized into three groups. The mean patient age was 66 years (range 60 to 74). Group 1 was given testosterone undecanoate 160 mg/day, the second group was given propionyl-L-carnitine 2 g/day plus acetyl-L-carnitine 2 g/day. The third group was given a placebo (starch). Drugs and placebo were given for 6 months. The assessed variables were total prostate-specific antigen, prostate volume, peak systolic velocity, end-diastolic velocity, resistive index of cavernosal penile arteries, nocturnal penile tumescence, total and free testosterone, prolactin, luteinizing hormone, International Index of Erectile Function score, Depression Melancholia Scale score, fatigue scale score, and incidence of side effects. The assessment was performed at intervals before, during, and after therapy. RESULTS: Testosterone and carnitines significantly improved the peak systolic velocity, end-diastolic velocity, resistive index, nocturnal penile tumescence,International Index of Erectile Function score, Depression Melancholia Scale score, and fatigue scale score. Carnitines proved significantly more active than testosterone in improving nocturnal penile tumescence and International Index of Erectile Function score. Testosterone significantly increased the prostate volume and free and total testosterone levels and significantly lowered serum luteinizing hormone; carnitines did not. No drug significantly modified prostate-specific antigen or prolactin. Carnitines and testosterone proved effective for as long as they were administered, with suspension provoking a reversal to baseline values. Only the group 1 prostate volume proved significantly greater than baseline 6 months after testosterone suspension. Placebo administration proved ineffective. Negligible side effects emerged. Prostate-specific antigen, prostate volume, nocturnal penile tumescence, total and free testosterone, prolactin, luteinizing hormone, erectile function scores, depression, fatigue and other parameters were assessed before treatment, at three months, andafter the treatment period. –Erectile function, sexual desire, sexual satisfaction and nocturnal penile tumescence increased over the course of the study in the group receiving testosterone as well as the group receiving carnitine. In addition, the group receiving carnitine experienced an increase in orgasm and general sexual well-being. Erectile function and nocturnal penile tumescence were significantly more improved in this group than in those receiving testosterone. While both treatments lowered depression scores, carnitine’s effect was greater. –Predictably, treatment with testosterone increased serum total and free testosterone”Click to Continue > by SaverExtension” and decreased luteinizing hormone levels, but carnitine’s effect on these hormones was not significant. Treatment with carnitine was not associated with an increase in prostate volume as was testosterone treatment after six months. PSA levels did not increase in any of the groups. –Fatigue was likewise improved inboth groups. Although both testosterone and carnitine improved symptoms of male aging, there was overall superior benefit to carnitine with no real side effects.
